چکیده
Basal cell carcinoma (BCC) is the most common cutaneous malignancy and characterized by sustained Hedgehog (Hh) signaling. Despite major insights into molecular basis of BCC, relatively little known about how BCC tumor cells affect other types in microenvironment. It has been frequently observed that a subset tumors are clinically pigmented. Using immunostaining for melanocyte-specific antigens, we found human mouse BCCs contain nodules with abundant proliferating melanocytes, even not grossly We performed single-cell RNA sequencing to search potential tumor-cell derived factors capable driving melanocyte proliferation. expression potent mitogens Endothelins 1 2 (Edn1, Edn2) was markedly upregulated cells, whereas corresponding Endothelin receptor type B (Ednrb) largely restricted melanocytes. Conditioned medium from BCC-like but squamous or normal keratinocytes, stimulated proliferation cultured Moreover, proliferative response melanocytes conditioned completely blocked small-molecule EDNRB inhibitor. Finally, biopsies obtained patient before during treatment Hh pathway inhibitor vismodegib, regression correlated reduced apoptosis tumor-associated Taken together, our data identify as frequent inhabitants microenvironment, point Edn1 and/or Edn2 likely tumor-cell-derived factor(s) may explain well-known poorly understood propensity pigmentation BCCs.

Pigmentation, Melanocyte Colonization, and p53 Status in Basal Cell Carcinoma
Basal cell carcinoma (BCC) is the most common neoplasm in the Caucasian population. Only a fraction of BCC exhibits pigmentation. Lack of melanocyte colonization has been suggested to be due to p53-inactivating mutations in the BCC cells interfering with the p53-proopiomelanocortin pathway and the production of alpha melanocyte-stimulating hormone in the tumor. To evaluate this, we determined t...
متن کاملExosomes released by keratinocytes modulate melanocyte pigmentation
Cells secrete extracellular vesicles (EVs), exosomes and microvesicles, which transfer proteins, lipids and RNAs to regulate recipient cell functions. Skin pigmentation relies on a tight dialogue between keratinocytes and melanocytes in the epidermis. Here we report that exosomes secreted by keratinocytes enhance melanin synthesis by increasing both the expression and activity of melanosomal pr...
متن کاملUpregulation of Endothelin-1/Endothelin A Receptor Expression Correlates with Heparanase Expression in Ovarian Carcinoma
Background: Heparanase and endothelin-1/endothelin A receptor (ET-1/ETAR) expressions increase in cancer. This condition enhances tumor progression and correlates with poor survival. Limited data are documented regarding the role of heparanase and ET-1/ETAR in epithelial ovarian cancer (EOC). We sought to characterize the correlation between heparanase and ET-1/ETAR in EOC.Methods: Thirty patie...
متن کاملThe biology of melanocyte and melanocyte stem cell.
The melanocyte stem cells of the hair follicle provide an attractive system for the study of the stem cells. Successful regeneration of a functional organ relies on the organized and timely orchestration of molecular events among distinct stem/progenitor cell populations. The stem cells are regulated by communication with their specialized microenvironment known as the niche. Despite remarkable...
متن کاملMast cells and histopathologic variants of basal cell carcinoma
Background and aim: The stroma of basal cell carcinoma (BCC) is rich in mast cells. Morpheaform BCC has been reported to contain much more mast cells than the other types of BCC, but their significance remains unknown. In this study we investigated the significance of mast cells related to BCC and possible relationship between increased number of mast cells and clinical and histological p...
